What is CTL assay?
Among measures of CMI, the cytotoxic T Lymphocyte (CTL) response is recognized as perhaps the most relevant functional measure that reflects cell-mediated acquired immune defense against viral infections and cancer. For this reason the CTL assay is an excellent candidate for evaluation of potential immunotoxicity.
What is a CTL T cell?
CD8+ T cells (often called cytotoxic T lymphocytes, or CTLs) are very important for immune defence against intracellular pathogens, including viruses and bacteria, and for tumour surveillance. This pore allows the granzymes also contained in the cytotoxic granules to enter the infected or malignant cell.
What do CTLs do?
CTLs are cytotoxic against tumor cells and host cells infected with intracellular pathogens (Fig. 4-7). These cells (1) express the CD8 coreceptor and (2) destroy infected cells in an antigen-specific manner that depends on the expression of MHC class I molecules on APCs.
What cell does perforin produce?
Normal Function Perforin is a major component of structures called cytolytic granules within T cells and NK cells. One of the main ways in which T cells and NK cells destroy other cells is to transport and secrete these cytolytic granules, which contain cell-killing proteins, onto the membranes of the target cells.
What is the role of perforin in killing of target cells by CTL?
Cytotoxic CD8 T cells carry out their killing function by releasing two types of preformed cytotoxic protein: the granzymes, which seem able to induce apoptosis in any type of target cell, and the pore-forming protein perforin, which punches holes in the target-cell membrane through which the granzymes can enter.
How is CTL generated?
Cytotoxic T lymphocytes (CTLs) are generated by immune activation of cytotoxic T cells (Tc cells). They are generally CD8+, which makes them MHC class I restricted. CTLs are able to eliminate most cells in the body since most nucleated cells express class I MHC molecules.
