What is BrdU used for?
Bromodeoxyuridine (BrdU) is a thymidine analog that incorporates DNA of dividing cells during the S-phase of the cell cycle. As such, BrdU is used for birth dating and monitoring cell proliferation.
How do you dissolve BrdU?
For a 100X stock solution, prepare BrdU at 40 mg/mL in 10 mM Tris-Cl (pH 7.6) in a 15-mL screw-cap conical tube. Place the solution in a waterbath at ~50ºC and occasionally vortex vigorously until the BrdU is completely dissolved.
What is BrdU antibody?
BrdU is a thymidine analogue and when offered to proliferating cells it is incroporated into reduplicating cells. The antibody is specific for DNA in which BrdU has been incorporated. In immunoassays this antibody reacts strongly with free or carrier-protein coupled BrdU but not with other nucleosides.
How do you use BrdU?
Dilute BrdU in PBS to make a sterile solution of 10 mg/mL. For mice, as a general rule, inject the BrdU solution to a concentration of 100 mg/kg….Oral administration
- Dilute BrdU to 0.8 mg/mL in drinking water.
- After treatment with BrdU, the animals can then be sacrificed according to standard protocols.
How do you reconstitute BrdU?
We usually used NaOH (0.02N) to dissolve BrdU powder. For a final concentration of 100 mg BrdU / kg of animal we dissolve 33mg of BrdU in 1 ml of NaOH (Stock solution).
Does BrdU dissolve in water?
BrdU takes up to a few hours to dissolve at 37oC depending on amount. 2. Prepare BrdU for supplementation in drinking water (if necessary). Make up solution of 0.8mg/mL BrdU in water (e.g. 0.240g in 300mL).
What is BrdU Labelling?
BrdU (Bromodeoxyuridine / 5-bromo-2′-deoxyuridine) is an analog of the nucleoside thymidine used in the BrdU assay to identify proliferating cells. Print this protocol. BrdU labeling can be performed in vitro for cell lines and primary cell cultures, or in vivo for labeling cells within a living animal.
